ASUSUpdate is an application that was not designed for your motherboard. You can't use it and you should avoid using it to prevent damage or make the motherboard unusable.

Yes, the application will work because this is an Asus motherboard. Install AsusUpdate and download the BIOS file automatically from the built-in download manager or from the official website for this motherboard.

Notice that BIOS upgrade can cause damage to your motherboard it it's wrongly performed. Try consulting the manual first and then perform the operation. If the computer is working just fine, BIOS update is not recommended.
