I am using Garmin Nüvi 255W. Can I create my own route with Garmin Express and then upload it to my Garmin device for use?

Garmin Express allows you to synchronize with Garmin Connect. No, it does not allow creating your own routes. You can do that with Garmin Connect and then synchronize the routes with your device through Garmin Express.
