I have a single license extension for version 8.5 that is installed on my Mac, and I have created a FDB collection on a local server. I would like to see if a Windows machine (which has access to the server) can now access this collection. Is this possible?

Yes. This is possible even if you are trying to access the collection that you created on a Mac computer from a Windows machine. It seems that Extensis Portfolio creates a cloud storage space that can be accessed by any mobile platform like iOS or Android and Windows or Mac machines.
