Help with Saving Images with Scale Bar and Calibration in SeBaView

asked by about Laxco SeBaView

Hello,

I am currently using SeBaView for image acquisition and I have some questions regarding saving images with calibration and scale bars.

I want to save my images while preserving the original calibration information (µm/pixel). However, when I open the saved TIFF images in ImageJ, the units appear as inches, not micrometers. Could you please advise the correct procedure to save images so that the calibration is preserved?

I would also like to add a scale bar to the images and save it together with the image. Could you provide instructions on how to do this properly in SeBaView so that the scale bar reflects the correct calibration?

I have tried saving in TIF format, but the calibration information is not preserved. Any guidance would be greatly appreciated.

Thank you very much for your support.

To address your question about saving images with the correct calibration and scale bar in Laxco SeBaView:

  1. Ensuring Calibration is Saved:

    • When you initially set up your image acquisition, make sure that the software settings explicitly save calibration data. This might be a checkbox or an option in the export settings.
    • If your software doesn't seem to embed the calibration information correctly, check the software documentation or settings for specific options related to exporting or saving with metadata.
  2. Adding a Scale Bar:

    • SeBaView should have an option to overlay a scale bar on the images before saving.
    • Ensure that the scale bar reflects the correct calibration units (µm/pixel). This often involves inputting or confirming the calibration settings in the software before adding the scale bar.
  3. Preserving Calibration in TIFF Format:

    • TIFF files support metadata, so the calibration information should be theoretically preserved. If ImageJ is showing inches, the calibration might not be saved correctly.
    • After saving the image, open it again in SeBaView to check if the calibration information is intact within the software. If it is not, there may be an issue with SeBaView preserving metadata that is compatible with other software like ImageJ.
  4. Correct Procedure:

    • Save the image directly from SeBaView after adding the scale bar and ensure the calibration checkboxes/options are enabled.
    • Verify the TIFF properties in SeBaView. If there's an export setting for preserving export metadata, ensure it’s activated.
  5. Additional Suggestions:

    • Consider checking for any SeBaView updates or patches that might address this issue.
    • Correspond with SeBaView’s technical support for detailed advice tailored to the latest software version.

These steps should help in properly saving images with the correct calibration and scale bars, ensuring they are preserved when opened in other software like ImageJ.

In SeBaView, enable the scale bar and calibration settings before saving. Then export the image using the built-in save or export option to preserve both the scale bar and calibrated measurements.
