Will this make my Galaxy Note 10.1 2014 edition read my external HD that is not formatted FAT32?

Yes, the application is able to mount and detect non-FAT32 drives (especially NTFS). Simply install the application and then start it and assign the drive letters while the drive is connected. However, root might be needed since the application will trigger some system commands.
