If I measure Lab & Lch values for a sample in 2 different instruments (spectrophotometers), will the values differ? If so, to what extent?

Yes, the values will different if those spectrophotometers weren't calibrated the same way. This means you will have to interpret different values. However, if you have the software installed for the instruments, I recommend performing a double calibration and then measure the values. Even a slightest calibration value has an impact over the final data.
